The TNF Receptors p55 and p75 Mediate Chemotaxis of PMN Induced by TNFα and a TNFα 36–62 Peptide
The present study was performed to examine whether residues 36-62 of TNFalpha contain the chemotactic domain of TNFalpha, and whether the p55 and p75 TNF receptors are involved in TNFalpha induced chemotaxis. The chemotactic effect of TNFalpha on PMN was inhibited by the mAbs Hrt-7b and Utr-1, against the p55 and p75 TNF receptors, respectively. Both receptors may therefore be required for medi...
متن کاملIntratumoral TNFα improves immunotherapy
Solid tumors are frequently resistant to immunotherapy. We demonstrated that low-dose tumor necrosis factorα (TNFα), when directly targeted to the tumor environment, exerts dual effects by improving vessel functionality and activating immune cells. This vascular remodeling in an inflammatory context enhances active immunotherapy and promotes tumor regression.
متن کاملDeviations in Circulating TNFα Levels and TNFα Production by Mononuclear Cells in Healthy Human Populations
OBJECTIVES Tumor necrosis factor alpha (TNFα) plays a pivotal role in the inflammatory host response. The serum-level of TNFα and the production of TNFα by lympho/monocytes, however, seem to show high individual variations. The goal of the present study was to investigate the variations and inducibility of TNFα-activity in two age-groups of healthy volunteers. METHODS Sixty elderly, healthy v...
متن کاملSoluble TNF-alpha receptors bind and neutralize over-expressed transmembrane TNF-alpha on macrophages, but do not inhibit its processing.
Tumor necrosis factor alpha (TNF-alpha) is initially synthesized as a type II integral membrane protein (transmembrane TNF-alpha) after macrophage activation. In this study we have investigated some aspects of the regulation of expression and biological activity of transmembrane TNF-alpha by both soluble TNF-alpha receptors (sTNF-alphaR) and inhibitors of TNF-alpha processing. We show, using th...
متن کاملResponse of pediatric uveitis to TNFα inhibitors
Purpose Non-infectious uveitis (“uveitis”) is an important cause of visual loss amongst children. Although TNFa inhibitors (anti-TNFa) are increasingly used to treat pediatric uveitis, the outcomes of treatment with these agents have been poorly described. We are studying the largest, and only multicenter, group of patients with pediatric uveitis in order to more precisely estimate the outcomes...
